Our research group „Allelic imbalance in cardiomyopathies” examines the influence of burst-like transcription and unequal expression of the two alleles of disease-associated genes in heterozygously transmitted cardiac disorders. We use human induced pluripotent stem cell derived cardiomyocytes (hiPSC-CMs) as model organism for our research. We analyze (allele specific) gene expression using RT-qPCR, RNA-Seq, spatial transcriptomics, and RNA-FISH and functional alterations (contractility and calcium handling) in order to gain insight into basic transcriptional mechanisms (“burst-like transcription”) as pathomechanism and to test potential therapeutic approaches.
